DNA‐Mediated Protein Shuttling between Coacervate‐Based Artificial Cells

چکیده

The regulation of protein uptake and secretion is crucial for (inter)cellular signaling. Mimicking these molecular events essential when engineering synthetic cellular systems. A first step towards achieving this goal obtaining control over the release proteins from cells in response to an external trigger. Herein, we have developed artificial cell that sequesters releases proteinaceous cargo upon addition a coded chemical signal: single-stranded DNA oligos (ssDNA) were employed independently localization set three different ssDNA-modified proteins. signal allows multiple iterations triggered release, amount rate sequential This signaling concept was furthermore used directionally transfer between two populations, providing novel directions lifelike communication pathways inside higher order (proto)cellular structures.

Optomechanical photon shuttling between photonic cavities.

Mechanical motion of photonic devices driven by optical forces provides a profound means of coupling between optical fields. The current focus of these optomechanical effects has been on cavity optomechanics systems in which co-localized optical and mechanical modes interact strongly to enable wave mixing between photons and phonons, and backaction cooling of mechanical modes. Alternatively, ex...

The POU protein Oct-6 is a nucleocytoplasmic shuttling protein

Like many POU domain proteins, Oct-6 plays important roles during vertebrate development. In accord with its function as a transcriptional regulator during neurogenesis and myelination, Oct-6 is predominantly found in the nucleus. Nuclear import is mediated by a nuclear localization signal at the N-terminal end of the POU homeodomain. Here we show, that Oct-6 in addition contains a nuclear expo...

Nucleocytoplasmic shuttling of bovine herpesvirus 1 UL47 protein in infected cells.

Previous studies with transfected cells have shown that the herpes simplex virus type 1 (HSV-1) and bovine herpesvirus 1 (BHV-1) UL47 proteins shuttle between the nucleus and the cytoplasm. HSV-1 UL47 has also been shown to bind RNA. Here we examine the BHV-1 UL47 protein in infected cells using a green fluorescent protein-UL47-expressing virus. We show that UL47 is detected in the nucleus earl...

Ca2+ shuttling between endoplasmic reticulum and mitochondria underlying Ca2+ oscillations.

Although many cell functions are regulated by Ca(2+) oscillations induced by a cyclic release of Ca(2+) from intracellular Ca(2+) stores, the pacemaker mechanism of Ca(2+) oscillations remains to be explained. Using green fluorescent protein-based Ca(2+) indicators that are targeted to intracellular Ca(2+) stores, the endoplasmic reticulum (ER) and mitochondria, we found that Ca(2+) shuttles be...

Shuttling of galectin-3 between the nucleus and cytoplasm.

In previous studies, we documented that galectin-3 (M(r) approximately 30,000) is a pre-mRNA splicing factor. Recently, galectin-3 was identified as a component of a nuclear and cytoplasmic complex, the survival of motor neuron complex, through its interaction with Gemin4. To test the possibility that galectin-3 may shuttle between the nucleus and the cytoplasm, human fibroblasts (LG-1) were fu...
